Great Runes are rare items in Elden Ring that only drop from the main bosses of the game. Great runes are not used like typical runes in the game and have to be restored before they have a purpose.
How to restore Great Runes?
To restore a great rune, you first will need to acquire one. The first Great Rune you will typically acquire is from Godric the Grafted. Upon defeating him he will drop his Great Rune. Each great rune you acquire will need to be taken to a specific Divine Tower to be restored. You can view which tower by reading the item's description.

Luckily, Godric's Divine Tower is just west of Stormveil Castle and can be accessed by heading west after entering through the main castle gates. Be wary of the giants along the path, you can fight them or run past and access the teleportation gate that will take you to the other side of the bridge. Once reaching the top you will be able to restore the great rune using a Rune Arch.
How to use a Great Rune in Elden Ring
To use a Great Rune in Elden Ring, all you need to do is equip it. Equipping the Great Rune will give you a passive buff. However, this Great Rune buff will only last until you die. After you die, you will need to reactivate the rune following the steps above.
How to equip Great Runes?
Each great rune will have its own unique effect. Upon restoring your first great rune you will have an additional option when resting at a site of grace, called Great Runes. This will allow you to equip a single great rune.
